The LOAs are numbered based on how many there have been, either since the first contract was signed, or since the most recent contract was signed and all prior LOAs were integrated.
So there's no apostrophe prior to the number like it was a year. Just LOA 72.

Isn't that what the joke is? The lego pilot is complaining about LOA 72 being a piece of garbage and that's why he voted no on the contract, but the hidden part of the joke is that they have gone through 72 LOAs since the last contract?

In terms of the actual RJ airframes being on mainline property that's unlikely.
However in terms of blocks hours or from a "scope choke" perspective a significant amount of scope has been recaptured and will continue to be recaptured in the form of increased 717/MD88/A319/A320 flying. This has been a big factor behind the record hiring currently happening at Delta and United.
I used to fly the CRJ into airports like AGS, CAK and SGF all day everyday. Now I'm flying the MD88/90 there.

I always thought that a German explaining humor was kind of like a Brit explaining dental hygiene or a Frenchman explaining calvary charges.
But just so you can be in on the joke too...
The high number of the LOA isn't the hidden part of the joke. It's pretty normal to have very high numbered LOAs as they don't normally reset the count with each new contract. Lots of things that are in LOAs (code shares etc) don't get integrated into the actual contract language when a new one is signed so the numbering system just keeps going. I've seen LOAs in the 100s before. So #72 doesn't actually imply that there have been 72 modifications since the last contract was signed but rather 72 modifications to the contract language, since they started keeping track of changes through LOA.
The teaching point I made to Hacker (mostly because he's planning on getting into the crazy 121 game some time and this falls under "nice to know" info) was that he referenced it as "LOA '72" which in English would be the same as saying "LOA 1972" as in, the LOA from 1972. Contracts (sometimes) get referenced that way as there probably is only one contract signed in a given year, but LOAs do not as there can be multiple ones signed in a year. The closest you'll get to that sort of notation might be something like LOA 15-1, LOA 15-2 etc but only a few places (Delta) do it that way.
The "hidden" joke, if there was one, was more about how the captain wouldn't say anything in response to the FO until he brought up work, work work, contract, work, work, work.
